Novel 10 kHz rapifleX Offers Breakthrough TOF/TOF Performance and Robustness for Primary Structure and PTM Determination in Proteomics, MALDI Tissuetyping and Biopharma Applications


At the 14th Human Proteome Organization World Congress (HUPO), Bruker today announced the new rapifleX MALDI-TOF/TOF mass spectrometer. MALDI-TOF/TOF is established as a powerful tandem mass spectrometry method for in-depth protein analysis. With its redesigned, next-generation TOF/TOF ion optics, the rapifleX system now offers significantly higher speed, mass resolution and mass accuracy. The rapifleX integrates a novel, 10 kHz smartbeam™ 3D scanning laser design for breakthrough mass-spec imaging capabilities with dramatically improved spatial resolution, image contrast and quality, and a new ion source for superior robustness and throughput in pathology research.

The ImageID™ workflow enables biomarker discovery in MALDI imaging cohort studies, with increased speed, resolution and mass accuracy with the rapifleX™ MALDI TOF/TOF. (Graphic: Business Wire) Stability and homogeneity of therapeutic proteins are of primary concern due to the effect on safety and efficacy. Undesired biochemical, structural and conformational forms can lead to a loss of potency or, in the worst-case, adverse side effects. As such, the evaluation of disulfide bonds is an important aspect in a comprehensive understanding of the chemical structure of a protein. Disulfide bond analysis is greatly enhanced by the better resolution and higher mass accuracy of the rapifleX™ MALDI-TOF/TOF. The analysis is fully automated and does not rely on a priori assumptions or target lists. The DisulfideDetect software is a quantum leap in ease-of-use for detecting and evaluating disulfide bonds in therapeutic proteins, such as monoclonal antibodies. It provides an easy to understand, visual workspace for disulfide bond analysis for both native and non-native bonds, as well as inter- and intra-chain disulfide bonds within the protein / antibody.

Tissue- and cell-type specific proteomics is a major advance for relevant biomarker discovery and identification, and requires MALDI tissue imaging studies which include the identification of detected biomarker candidates for validation and the subsequent development of targeted assays. The rapifleX MALDI-TOF/TOF extends the MALDI Tissuetyper® imaging capabilities with rapid, spatially resolved MS/MS data. It supports the patented ImageID™ workflow for biomarker discovery in MALDI imaging cohort studies by obtaining matching identifications of tryptic peptides in bottom-up imaging studies.
