Waters announced today it will introduce a new nanoflow chromatography system at ASMS, June 5 - 9, in San Antonio, TX. Waters® nanoACQUITY Ultra Performance LC System greatly enhances protein identification and characterization. Its new features increase the amount of information obtained from the most complex mixtures in proteomics applications.
The new suite of capabilities includes a unique direct nanoflow Ultra Performance LC approach, fully automated on-line two-dimensional liquid chromatography, a new fraction collector for LC/MALDI™ analysis and off-line two-dimensional LC applications. The Waters nanoACQUITY UPLC system now allows scientists to attain unparalleled analytical flexibility, sensitivity, reproducibility and resolution for protein identification and characterization.
Benefits of Ultra Performance LC Now Applied to NanoFlow Applications
Waters nanoACQUITY UPLC System is a first-of-its kind LC system designed to take full advantage of the chromatographic potential of novel sub two-micron particles at nanoscale flow rates to significantly enhance the resolution and peak capacity of a single chromatographic run, thereby providing more information per unit of time than any of today's nanoscale HPLC systems. To leverage the full potential of Waters' patented pressure-tolerant 1.7 mm reversed-phase particle technology, Waters' nanoACQUITY UPLC columns are now available in new lengths and capable of operating at pressures of up to 10,000 psi and at flow rates down to 200nl/min. to provide 2X the peak capacity of HPLC and unparalleled run-to-run reproducibility (<0.25 min relative standard deviation).
The combination of the nanoACQUITY UPLC with the enhanced speed, resolution and dynamic range of Waters Micromass ® Premier family of orthogonal time-of-flight (oa-Tof) mass spectrometers provide the highest LC/MS and MS/MS performance demanded of today's instruments for proteomics applications.
Adding Dimension to Protein Identification
A common approach for increasing the amount of information from complex proteomics samples is to reduce their complexity and dynamic range prior to mass spectrometry (MS) analysis by using two-dimensional liquid chromatography. To further improve the identification and increase the sequence coverage of high and low abundance proteins from complex samples, the Waters' nanoACQUITY UPLC system can now be configured for fully automated on-line 2D LC separations of complex protein digest samples using an ion exchange column in the first dimension, and reversed phase HPLC (nanoACQUITY columns with Atlantis ® chemistry) or UPLC (nanoACQUITY columns with BEH Technology™) in the second dimension. This easy-to-use system combines both high performance 2DLC and UPLC separations with minimal band broadening to vastly increase the information gained in the qualitative analysis of complex protein samples.
Maximizing Protein Identification by Combining UPLC, MALDI and Electrospray Ionization
For some of the most complex proteomics samples, many researchers choose to minimize sample complexity and dynamic range by first collecting discrete fractions from a gradient separation prior to MALDI or LC/ESI MS/MS analysis. In an agreement with CTC Analytics AG ( Zwingen, Switzerland), the CTC PAL™ microfraction collection platform will be sold with Waters nanoACQUITY UPLC system (utilizing HPLC and UPLC separations) for time-based fraction collection onto MALDI targets (96 or 384 format) or into 96-well or 384-well microtiter plates for LC/electrospray ionization (ESI) analysis. The popular CTC PAL MALDI Spotter/Fraction Collector is easy to use, minimizes band broadening to preserve high peak capacity nanoscale and capillary separations (using 300 m m to 75 m m ID columns), and is compatible with both HPLC and UPLC-based separations. By automating matrix addition to sample fractions, the CTC PAL MALDI Spotter/Fraction Collector can enable direct MALDI analysis and supports all MALDI target plates employed by the Micromass MALDI micro MX TM and new MALDI-enabled Q-Tof Premier TM Mass Spectrometer introduced at Pittcon 2005. The combination of the nanoACQUITY UPLC system and the flexible CTC PAL MALDI Spotter/Fraction Collector enables researchers to perform the best off-line nanoscale separations possible and, in combination with MALDI and electrospray mass spectrometry, complementary techniques that maximize the information content from every sample.
