Shimadzu Corporation announces the release of its Quick-DB GC/MS/MS forensic toxicological database (Quick-DB Forensic) for triple quadrupole gas chromatograph mass spectrometers (GC-MS/MS). This product was developed in cooperation with the Department of Legal Medicine and Bioethics, Postgraduate School of Medicine (Associate Professor Kei Zaitsu and Assistant Professor Maiko Kusano), Nagoya University.

This database includes all the information necessary for analyzing the 68 toxicological substances often involved in drug poisoning cases in Japan, including optimal pretreatment methods, analytical conditions (GC/MS/MS measurement parameters and calibration curves), and data analysis parameters. Therefore, it provides a total solution for trace analysis of toxicological substances contained in blood.

Background to the Development

Departments of forensic medicine at universities, which perform toxicological analysis in autopsies, and emergency hospitals that must quickly identify substances responsible for poisonings, reportedly are facing a shortage of staff skilled in analysis. Therefore, there is demand for a system that allows users with minimal experience to easily perform all steps, from pretreatment to data analysis, and output final results at a given level. Another issue is the cost and labor necessary for purchasing the many standard samples required for quantitation of toxicological substances and managing them in accordance with given laws and regulations.

To meet these demands, the Quick-DB GC/MS/MS forensic toxicological database (Quick-DB Forensic) incorporates all the analytical technology and pretreatment expertise accumulated by the Department of Legal Medicine and Bioethics, Postgraduate School of Medicine, Nagoya University, so that even a user with limited analytical experience can easily obtain accurate analytical results without using a standard sample.


1. Providing a Total Solution for the Analysis of Toxicological Substances

The database includes information on pretreatment, sample analysis, and data analysis for 68 toxicological substances commonly found in drug poisoning cases. When used in combination with a Shimadzu GCMS-TQ series system, it provides a total solution for toxicology analysis. It uses the simple QuEChERS method for pretreatment, enabling inexperienced users to pretreat samples easily. The database contains not only optimized MRM transitions and other analysis conditions, but also retention indices, calibration curve information, and other data analysis conditions. Therefore, analysis methods can be created easily via the automatic method creation function (Smart MRM).

2. Achieves Analysis Without Using Any Standards

To improve the accuracy of simplified quantitative analysis when not using a standard sample, the selection of the internal standard and the pretreatment recovery rate must be considered. In this database, six deuterium-labeled toxicological substances with the same behavior as the registered compounds are used as the internal standards. In addition, it uses calibration curve information prepared by pretreating whole blood samples spiked with a standard sample. This reduces quantitative errors resulting from matrix interference and pretreatment recovery rates, which can have a significant effect on quantitative accuracy. Consequently, accurate analysis results without standards is obtainable.

3. Smart Scheduling Function Automatically Injects Blanks

When target compounds are detected in test samples at high concentrations, false positives may occur in the next sample due to carryover within the system. In such cases, results are confirmed by measuring the test sample again. If sample volumes are small, however, pretreatment will need to be repeated, significantly reducing operating efficiency. With this database, when target compounds are detected in test samples at a higher than prescribed concentration, the risk of false positives due to carryover can be avoided because the system is equipped with a Smart Scheduling function, which measures solvent blanks automatically.
